The pycA was selected as a barcode gene that has the potential to identify Bacillus cereus group species and has stronger species discrimination than the 16S rRNA gene. The purpose of this study was to reveal species nitrogen-fixing (B2), Indole-3-Acetic Acid (IAA) producing (PIS), and cellulolytic (PSS) of Bacillus cereus group from Ranu Pani based on the pycA gene. This research was conducted by isolating bacterial DNA, continued with amplifying the pycA gene sequence using forward primer pycA-F (5'-GCG TTA GGT GGA AAC GAA AG-3') and reverse primer pycA-R (5'-CGC GTC CAA GTT TAT GGA AT-3'). Amplified target genes sequence were sequenced at 1st BASE Laboratories Malaysia. Phylogenetic analysis and genetic distance were carried out using MEGA-X software. Based on phylogenetic tree reconstruction and genetic distance analysis, we suggest that samples B2 and PIS were Bacillus cereus, Bacillus thuringiensis, or Bacillus anthracis (genetic distance 0.004). The PSS sample was suspected as Bacillus cereus or Bacillus tropicus (genetic distance 0.002). Based on the results, it was known that the B2, PIS, and PSS isolates were still biased. Further identification using whole genome-based analysis is needed to determine the exact species to which our bacteria belong. © 2023 American Institute of Physics Inc.. All rights reserved.
