Endophytic fungi isolated from yellow champaca (Michelia champaca L.) plant: a histological observation, identification, and secondary metabolite analysis

Abstract

Endophytic fungi live in some plant tissues, included the yellow champaca tissues. This fungi produce secondary metabolites that could protect the host plant from the pathogenic bacteria. This research aimed: to determine the endophytic fungi hyphae position in leaves, twigs, and flower petal in M. champaca L. plant tissues by histological observation; to identify endophytic fungi isolated from M. champaca L., and to analyze several secondary metabolite contents produced from each endophytic fungi species. The plant parts were prepared for the histological observation. The endophytic fungi were isolated by inoculating the plant parts on a PDA plate medium and incubated in 27 ℃ for a week. Each endophytic fungi culture was cut into 5 x 1 cm in size and inoculated on Potato Dextrose Broth (PDB) medium for analyzinge secondary metabolites content by using the spectrophotometric methods. The research result were the endophytic fungi hyphae was found in the leaves epidermal cell wall, stomata neighbor cell wall, on the twig parenchyma and epidermal cell wall, and petal epidermal cell wall., Eleven endophytic fungi species were found in leaves, twigs, and flower petal of M. champaca., The secondary metabolite produced by each endophytic fungi were consist of flavonoid, alkaloids, tannins, saponins, and terpenoids with different contents. © 2024 American Institute of Physics Inc..
