DNA Isolation and Amplification of the Cytochrome b Gene from Drosophila melanogaster

Abstract

Drosophila melanogaster is the most popular model organism in the field of Biology. Unfortunately, in Indonesia, the utilization of this organism is still far from optimal. This study aimed to isolate DNA and amplify the cytochrome b gene of D. melanogaster. The stages of the research were 1) culturing D. melanogaster; 2) sample treatment process; 3) DNA extraction and purification; 4) test the results of DNA extraction; 5) cytochrome b gene amplification; and 6) analysis of cytochrome b gene amplification results. G-Spin TM Total DNA Extraction kit successfully isolated the genome of D. melanogaster. In a thermo-cycler that was set with a lid temperature of 104°C, pre denaturation 94°C for 5 minutes, denaturation 94°C for 1 minute, annealing for 1 minute, elongation 72°C for 2 minutes, and post elongation 72°C for 5 minutes, the cytochrome b gene was amplified at annealing temperatures of 57°C and 58°C. These findings are expected to initiate and encourage wider use of D. melanogaster in various research fields in Indonesia, particularly in molecular studies. © 2025 American Institute of Physics Inc..
